Before you begin

• Open the current ASCP BOC content guideline for the credential and testing period.
• List three strongest and three weakest content areas.
• Choose fixed study blocks on your calendar.
• Create a missed-question log: topic, why missed, correct rule, next review date.
• Gather a primary textbook and laboratory examples/SOPs for comparison.

Week 1 — Fixation and preanalytics

• Complete the Fixation guide through preanalytics.
• Build a table of major fixatives: mechanism, best use, limitation, pigment.
• Review IHC/ISH effects of fixation and decalcification.
• Complete the Fixation quiz and review every explanation.
• Use the quick card and NBF worksheet for closed-book recall.

Week 2 — Processing and decalcification

• Map dehydration → clearing → infiltration and reagent compatibility.
• Compare biopsy, routine, fatty, fibrous, and rapid schedules.
• Compare strong acid, weak acid, and EDTA decalcification.
• Practice artifact-to-cause troubleshooting.
• Complete the quiz and create five “if/then” processing rules.

Week 3 — Embedding, microtomy, and cryostat

• Practice orientation rules for skin, GI, tubular structures, cysts, node, muscle, kidney, and biopsies.
• Review blade, angle, temperature, speed, thickness, and water-bath interactions.
• Create an artifact matrix for compression, chatter, lines, folds, holes, lifting, and floaters.
• Review cryostat temperature, safety, decontamination, and molecular contamination.
• Complete quiz and use the orientation/artifact PDFs.

Week 4 — Routine H&E and Special Stains

• Write the full H&E sequence from memory.
• Explain progressive/regressive staining, differentiation, bluing, eosin pH, and dehydration.
• Build a special-stain grid: target, chemistry, color, control, critical step.
• Practice PAS, trichrome, reticulin, VVG, Congo red, GMS, AFB, iron, Von Kossa, and Fontana concepts.
• Complete both quizzes and review control failures.

Week 5 — Laboratory operations

• Distinguish QC, QA, quality improvement, validation, verification, and calibration.
• Review maintenance and controls for each major histology instrument.
• Study the hierarchy of controls and common chemical, biologic, sharps, heat, and ergonomic hazards.
• Write a sample deviation, containment, root cause, CAPA, and effectiveness check.
• Complete the quiz and review local examples.

Open Laboratory Operations

Week 6 — HTL extension and integration

• Complete IHC/ISH fundamentals: controls, retrieval, detection, and preanalytics.
• Review validation and lot-bridging logic.
• Complete cumulative practice without notes.
• Rank missed questions by content area and root cause.
• Repeat weak modules and explain all previously missed questions aloud.

Exam-week approach

• Use the official content guideline to confirm no major topic has been skipped.
• Review concise tables and missed-question rules rather than rereading everything.
• Practice choosing the best answer, not merely a technically possible answer.
• Watch for questions that begin with specimen type, downstream assay, artifact, or control result.
• Do not change well-established answers based only on anxiety; change when you identify a concrete reasoning error.
• Protect sleep and logistics. Preparation quality falls when the final days become an all-night cram.
